Number plates - Anyway to Guarantee you get them??

VERY interested in a few personalised plates I have in mind; some are released next year and some the following. Obviously they give rough release dates. Is there any way of guaranteeing I can get them and not get beaten to it by a reg. company? Or at least how do I give myself the best opportunity to get them? Or is it just a case of whoever presses 'buy now' first on the dvla site?
Also, when you buy a new plate does it have to go straight on to a car or can you buy it and put it on retention straight away?
