The first question is: Do you actually use a VPN?
If not, it's some buttmunch's idea for fun, so ignore it. Your computer won't tell you that your VPN has been directed anywhere, as IP address translation happens completely transparently to the user, and in most cases, the operating system.
If you do have a VPN connection, delete the connection and recreate it, ensuring that you enter the correct VPN server's hostname or IP address

if you don't know if you have one, chances are you don't.
VPNs are used for connecting to other people's networks from remote locations, for example, using your work computer whilst sat at your home computer.
Block it and see what stops working - you'll soon find out if its something important, and then do all the ususal - check your AV and anti-spyware are up to date, then do a full scan to make sure you've not caught any nasties
After a bit of digging, that IP address appears to be located in India: http://wq.apnic.net/apnic-bin/whois....203.163.243.40
Unless you know anyone in India that wants to use your computer with your permission, block it, scan it, clean it

bloomin thing is back
virtual private network traffic has been detected to 203.163.243.40
do you want to automatically set a rule to allow vpn communications with this server
checked everything, and found nothing...

First off, NO you DON'T want to allow it.
Don't worry, you're not gonna have to wipe it and start again!
One more check that your computer's clean:
Goto your Network Connections control panel - you should have probably 3 icons in there tops: Make New Connection, Local Area Connection and your internet connection (which may also be called 'Local Area Connection' ) if you dial one with a modem.
Anything else, like 'Porn dialler', or 'XXX Boobs' etc can be safely binned.
Next, restart your computer in Safe Mode and get your anti-crap tools out - do a scan while in Safe Mode as this prevents the bastard programs from starting in the first place.
Whilst its scanning, have a beer and some grub then check your Startup Group ( Start -> All Programs -> Startup) and remove anything you don't recognise.
VPN connections don't just happen, unless someone at the other end is trying to connect to your computer thinking its a VPN server, or hoping its a VPN server that they can use to hack into a network. Either way - if you didn't start it, always block it unless you know what it is.
