Maybe a silly question but, do I need the ignition on when doing this

Also, the resovoir was tottally empty I noticed since removing all the brake lines for the underside, I have filled it up and pumped the pedal a few times but have nothing, will it need a lot of pumps to get the fluid back round?

p.s resouvoiuris not empty again still just under max
