You'd be as well to have the lump tweaked once back in the car too, to check/adjust for any differences.

A dyno basically allows load to be placed on the motor under precisely monitored and controlled conditions, and has it's own chargecooler. The main differences (usually) to take into account are IC, exhaust and your wiring loom (if it has any issues for example).
