The problem you are having is that the metering flap is not being displaced by the airflow going into the engine.

Have someone depress the metering flap slightly (Less than 1mm) whilst you crank her with WOT and see if she fires... If she does you are either leaking air into the engine or the base settings are no longer suitable for your engine and she needs setting up to make her run.
